備考 | The ALSV vector (RNA2) has been engineered to create a cloning site for introduction of gene fragments to induce gene silencing.
The Apple latent spherical virus (ALSV) gene silencing vector will be used to test gene function in cucumber and melon plants. Sequence fragments of genes to be silenced will be inserted into the vector cloning site in pEALSR2LR5 in the ALSV RNA2 as described on Igarashi et al. 2009 (Virology 286:407-416).
The ALSV VIGS vectors (RNA1 and RNA2) will be amplified in E. coli, plasmids purified, and mechanically inoculated onto leaves of Chenopdium quinoa in the growth chamber. To test for gene silencing infected C. quinoa leaves will be used to mechanically inoculate cucumber or melon in the growth chamber or greenhouse.
ALSV is a useful silencing vector because it does not cause symptoms or disease, including on apple (Li et al., 2004, Archives Virology 149:1541-1558).
Although ALSV can experimentally infect a relatively wide range of plant species, natural infection of ALSV has only been documented in one apple tree in Morioka, Japan and did not spread in the orchard over a period of more than two decades (Nakamura et al., 2011., J Gen Plant Pathol: 7748-53).
The virus did not have any obvious symptoms in apple trees inoculate by grafting. No other natural hosts have been identified.
